Choosing the right preparation type for Agrobacterium strains (such as GV3101, EHA105, or AGL-1) depends on your specific plant transformation workflow:
1. Chemically Competent Cells Method:
These cells take up DNA through a heat-shock method.
Usage: They are convenient and do not require specialized equipement, though they generally yield lower transformation efficiencies compared to electroporation.
20-50 mikroliter of chemically competent cells are thawed on ice, mixed with 10 ng of plasmid DNA, heat-shocked (usually in a water bath), and then incubated in an antibiotic-free recovery medium before being plated.
2. Electrocompetent Cells Method:
These cells take up DNA using an electrical pulse (electroporation).
Usage: This is widely considered the most effective method for Agrobacterium transformation. It is highly recommended if you are introducing large plant binary vectors or creating cDNA/gDNA libraries.
Workflow: Cells are gently thawed on ice, mixed with DNA, and transferred to a chilled electroporation cuvette. They are subjected to a high-voltage pulse (e.g., field strengths of 11-18 kV/cm.
Afterward, recovery medium is added immediately to help the cells recover.
Key Handling Tips for Agrobacterium strains
Temperature Sensitivity:
Agrobacterium cells are highly sensitive to temperature changes and mechanical lysis. Always thaw them completely on wet ice and handle gently.
Mixing:
Never vortex competent cells. Always mix cell/DNA suspensions by gently tapping the tube.
Antibiotic Selection:
Ensure your growth and selection media include the appropriate resistance markers for the specific strain (e.g., Rifampicin for EHA105 or Gentamycin for GV3101).
